应用笔记：This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human FETUB. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human FETUB and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human FETUB, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human FETUB. You can calculate the concentration of Human FETUB in the samples by comparing the OD of the samples to the standard curve.

结果的对照，因此对照品，特别是阳性对照品的基本组成应尽量与检测标本的组成相一致。以人血清为标本的测定，对照品最好也为人血清，因为正常人血清在各种ELISA模式中可产生不同程度的本底。由于大量正常人血甭较难得到，国外试剂盒中的对照品多以复钙人血浆(recalcified human plasma)为原料，即在血浆中加入钙离子，使其中的纤维蛋白质凝固，除去凝块后所得的液体，其组成与血清相似。阴性对照品须先行检测，确定其中不含待测物质。例如HBsAg检测的阴性对照品中不可含HBsAg，最好抗HBs也是阴性
