| 背景资料 | The protein encoded by this gene is a member of the LAR protein-tyrosine phosphatase-interacting protein (liprin) family. Liprins interact with members of LAR family of transmembrane protein tyrosine phosphatases, which are known to be important for axon guidance and mammary gland development. It has been proposed that liprins are multivalent proteins that form complex structures and act as scaffolds for the recruitment and anchoring of LAR family of tyrosine phosphatases. This protein has been shown to bind the calcium/calmodulin-dependent serine protein kinase (MAGUK family) protein (also known as CASK) and proposed to regulate higher-order brain functions in mammals. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-PPFIA2 antigen affinity purified polyclonal antibody (A11773) and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for PPFIA2 at approximately 143 kDa. PPFIA2 was detected in a paraffin-embedded section of rat brain tissue. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB (Catalog # AR1022) as the chromogen.|Figure 3. Overlay histogram showing U251 cells stained with A11773 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-PPFIA2 Antibody (A11773, 1 μg/1x106 cells). DyLight?488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody. Isotype control antibody (Green line) was rabbit IgG (Catalog # BA1045) (1 μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.[/list_product_images]
