| 背景资料 | N-terminal EF-hand calcium-binding protein 3 is a protein that in humans is encoded by the NECAB3 gene. The protein encoded by this gene interacts with the amino-terminal domain of the neuron-specific X11-like protein (X11L), inhibits the association of X11L with amyloid precursor protein through a non-competitive mechanism, and abolishes the suppression of beta-amyloid production by X11L. This protein, together with X11L, may play an important role in the regulatory system of amyloid precursor protein metabolism and beta-amyloid generation. The protein is phosphorylated by NIMA-related expressed kinase 2, and localizes to the Golgi apparatus. Multiple transcript variants encoding different isoforms have been found for this gene. |

After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-NECAB3 antigen affinity purified polyclonal antibody (A11525-3) and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for NECAB3 at approximately 44 kDa. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-NECAB3 Antibody (A11525-3, 1 μg/1x106 cells). DyLight?488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody. Isotype control antibody (Green line) was rabbit IgG (Catalog # BA1045) (1 μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.[/list_product_images]
