| 背景资料 | Protein NDRG4 is a protein that in humans is encoded by the NDRG4 gene. This gene is a member of the N-myc downregulated gene family which belongs to the alpha/beta hydrolase superfamily. The protein encoded by this gene is a cytoplasmic protein that is required for cell cycle progression and survival in primary astrocytes and may be involved in the regulation of mitogenic signalling in vascular smooth muscles cells. Alternative splicing results in multiple transcripts encoding different isoforms. |

After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-NDRG4 antigen affinity purified polyclonal antibody (A04972-1) and probed with a goat anti-rabbit IgG-HRP secondary antibody (Catalog # BA1054). The signal is developed using ECL Plus Western Blotting Substrate (Catalog # AR1197). A specific band was detected for NDRG4 at approximately 37 kDa. NDRG4 was detected in an immunocytochemical section of U2OS cells. Cy3-conjugated Anti-rabbit IgG Secondary Antibody (red)(Catalog#BA1032) and Dylight488-conjugated Anti-mouse IgG Secondary Antibody (Green) (Catalog # BA1126) were used as secondary antibody. |Figure 3. Overlay histogram showing 293T cells stained with A04972-1 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-NDRG4 Antibody (A04972-1, 1 μg/1x106 cells). DyLight?488 conjugated goat anti-rabbit IgG (BA1127, 5-10 μg/1x106 cells) was used as secondary antibody. Isotype control antibody (Green line) was rabbit IgG (Catalog # BA1045) (1 μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.[/list_product_images]
