| 背景资料 | Alpha-2-HS-glycoprotein (AHSG) also known as fetuin-A is a protein that in humans is encoded by the AHSG gene. Fetuin-A belongs to the fetuin class of plasma binding proteins and is more abundant in fetal than adult blood. This gene is mapped to 3q27. Fetuin-A is a secreted partially phosphorylated glycoprotein with complex proeolytic processing that circulates in blood and extracellular fluids. The protein is commonly present in the cortical plate of the immature cerebral cortex and bone marrow hemopoietic matrix. It is involved in several functions, such as endocytosis, brain development and the formation of bone tissue. Fetuin-A is also a potent inhibitor of pathological calcification. |

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.Lane 1: Recombinant Mouse FetuinA Protein 0.5ng After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FetuinA antigen affinity purified polyclonal antibody (Catalog # PB0242) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FetuinA at approximately 58KD. The expected band size for FetuinA is at 58KD.[/list_product_images]
