phi29 DNA Polymerase，即phi29 DNA聚合酶，是一种重组表达纯化的源于枯草芽孢杆菌Bacillus subtilis噬菌体phi29的DNA聚合酶。phi29 DNA聚合酶具有DNA链置换 (strand displacement) 活性和很强的DNA连续合成能力(processive synthesis property)，可以合成超过70kb的长度，常用于在体外进行不依赖于热循环的等温DNA扩增(isothermal DNA amplification，也常简称为等温扩增)。
phi29 DNA Polymerase具有很强的链亲和力，单次聚合反应可以实现长度超过70kb的连续聚合延伸，从而适合用于对全基因组进行稳定扩增。phi29 DNA Polymerase虽然可以扩增线状或者环状的双链DNA，但其底物倾向于是单链DNA或单链RNA。phi29 DNA Polymerase具有3'→5'外切酶活性，因此可保证扩增反应的高保真性。
用途：phi29 DNA Polymerase可用于高保真等温PCR、滚环复制(Rolling Circle Replication，RCA)，多重置换扩增(Multiple Displacement Amplification，MDA)和单细胞、病原微生物以及宏基因组(metagenome)的全基因组扩增(Whole Genome Amplification，WGA)、干血斑样本扩增DNA、单细胞基因组扩增、SNP基因分型和STR/微卫星分析等。phi29 DNA Polymerase还可应用于protein-primed DNA扩增、RNA-primed DNA扩增，以及致死基因的无细胞体系的克隆等相关实验。
