背景资料:KLF4 Monoclonal Antibody: KLF4 is a transcription factor that functions as both a transcriptional activator and repressor to regulate proliferation and differentiation of multiple cell types. The role of KLF4 in embryonic development suggested that it might be useful in the creation of stem cells that might be useful in cell replacement therapies in the treatment of several degenerative diseases. Artificial stem cells, termed induced pluripotent stem (iPS) cells, can be created by expressing KLF4 and the transcription factors POU5F1, Sox2, and Lin28 along with c-Myc in mouse fibroblasts. More recently, experiments have demonstrated that iPS cells could be generated using expression plasmids expressing KLF4, Sox2, POU5F1 and c-Myc, eliminating the need for virus introduction, thereby addressing a safety concern for potential use of iPS cells in regenerative medicine. KLF4 interacts directly with POU5F1 and Sox2 in iPS and ES cells and activates the target gene NANOG.

细胞和嗜性粒细胞,使之释放组胺,引起血管扩张,通透性增加,平滑肌收缩及局部水肿。但其作用远较C5a弱。此外,C3a还具有使吞噬细胞定向移动以促进吞噬的趋化作用,以及抑制特异抗体反应、非特异性多克隆反应和抑制白细胞移动抑制因子(LIF)产生的作用。C3b的生物学活性�广,概括起来有以下几个方面:(1)参与替代途径中两种C3转化酶[起始C3转化酶(C3bB)和放大C3转化酶(C3bBb)],以及两条途径中两种C5转化酶(C4b2a3b和C3bnBb)的形成;(2)启动替代激活途径中的正反馈放大回路
C3是补体二条激活途径的关键分子,C3活化后裂解为C3a、C3b两个片段,C3a很快被血清羧肽酶N裂解为C3adesarg,C3b被H、I因子灭活为C3bi,经蛋白酶水解为C3c、C3d、C3dg等,检测这些裂解产物即可反映C3的活化程度。C3a的定量检测采用竞争性抑制放射免疫法。其原理为用“I标记C3(I-C3)”与待测样本、兔抗人C3a多克隆抗体一起孵育,待测样本中的C3a(未标记抗原)能竞争性地与抗C3a结合,使C3a复合物(B)增多,而“I-C3”游离出来(F),根据B%来判断待测样本
