UPLC conditions
Chromatography was performed on an UPLC system with an autosampler at 4℃. The separation was carried out on an C18 column (1.7μm, 2.1 × 100 mm). The column temperature was maintained at 45℃. The analysis was performed with gradient elution using (A) acetonitrile, and (B) water with 0.1% formic acid as the mobile phase: 0–8 min, 1%–20% A; 8–11 min, 20%–30% A; 11–13 min, 30% –50% A; 13–16 min, 50%–99% A; 16–18 min, maintain 99% A; 18–19 min, 99%–1% A; held at 1% A for 3 min for equilibration of the column. Flow rate was set at 0.4 mL/min. The sample injection volume was 1μL. Throughout the analysis, all samples were kept at 4℃.
Mass spectrometry conditions
Mass Spectrometerwas connected to the UPLC system via electrospray ionization (ESI) interface. The capillary voltage and sample cone voltage, source offset voltage and the source temperature of positive and negativemodes were 2.5 kV, 60V, 80V, 120℃ and 2.0 kV, 40V, 60V, 130℃, respectively. The temperature of desolvation was 500℃. The cone and desolvation gas flow rates were 50 L/h and 800 L/h, respectively. The data acquisition rate was set at 0.2 s/scan, with a 0.1 s (0.02 interscan time) interscan delay. MS data was collected in centroid mode from 50 to 1200 Da. For accurate mass acquisition, a lock-mass ofleucine enkephalin was used to ensure accuracy through the whole MS analysis.
