HIF1A 分子靶点综述
低氧诱导转录因子1α(HIF-1α/HIF1A)是转录因子异二聚体HIF-1α/β的调控成员。作为对缺氧的适应性反应的主要转录调节因子,HIF-1α在低氧条件下,与许多基因启动子中的缺氧反应元件(HREs)结合,激活40多个基因的转录,包括促红细胞生成素、葡萄糖转运蛋白、糖酵解酶、血管内皮生长因子、HILPDA和其他其蛋白产物增加氧输送或促进代谢对低氧适应的基因。HIF-1α还在胚胎血管生成、肿瘤血管生成和缺血性疾病的病理生理中起重要作用以及参与缺氧时神经元内线粒体的轴突分布和运输。
HIF-1α普遍存在于人和哺乳动物细胞内,常氧下(21% O2)也有表达,但合成的HIF-1α蛋白很快被细胞内氧依赖性泛素蛋白酶降解途径所降解,只有在缺氧条件下HIF-1α才可稳定表达。
