应用:
乙二.胺四乙酸二钠盐二水合物用于植物物种的种子萌发试验以及从苔藓Physcomitrella paten中提取蛋白质。 还用于裂解和液泡缓冲液,用于从矮牵牛花瓣中分离液泡。
二价阳离子的螯合剂。 对需要二价阳离子发挥活性的酶,比如金属蛋白酶具有抑制作用。
EDTA是金属蛋白酶和金属活化蛋白酶的抑制剂,有效浓度为1-10mM。该产物在金属蛋白酶的活性位点中充当锌离子的螯合剂,但EDTA还可以抑制其他金属离子依赖性蛋白酶,例如钙依赖性半胱氨酸蛋白酶。
EDTA可能干扰依赖于金属的生物过程。
作为抗凝剂,EDTA的二钠盐或三钾盐是最常用的。

从2%EDTA溶液中,每毫升全血可滴1-2滴用作抗凝血剂。
准备说明:
该产品在室温下缓慢溶于水,最高可达0.26M,约96 mg in最终体积为1ml。
该溶液的pH值将在4-6的范围内。
EDTA盐更多当pH值增加时可溶于水:盐形式的EDTA越多水溶液,因此,室温下的溶解度越高。
这可以通过向EDTA溶液中逐渐加入浓氢氧化.钠溶液。
储存/稳定性:
0.5M EDTA在水中(pH 8.5)的储备溶液在4°C下可稳定数月。EDTA溶液可能高压灭菌。
乙二.胺四乙酸二钠说明:二价阳离子的螯合剂,能抑制酶的活性,例如金属蛋白酶,因为金属蛋白酶需要二价阳离子的存在才有活性。
0.5M EDTA(pH=8.0)的配制:
称取EDTA-Na2.2H2O186.1g溶于800mL体积纯水中,加入19g氢氧化.钠,然后用NaOH调节pH至8.0,后定容。高压灭菌后使用。
注意事项:
(1)EDTA-Na2.2H2O不易溶于水,只有在碱性条件下才能溶解,pH调至8时才能够完全溶解。
(2)注意调节终体积溶液pH为8.0。
Application:
Ethylenediaminetetraacetic acid disodium salt dihydrate is used for seed germination experiments of plant species and protein extraction from moss Physcomitrella paten. It is also used for lysis and vacuole buffer, for separating vacuoles from petunia petals.
A chelating agent for divalent cations. It has inhibitory effects on enzymes that require divalent cations to exert activity, such as metalloproteinases.
EDTA is an inhibitor of metalloproteinases and metalloproteinases, with an effective concentration of 1-10mM. This product acts as a chelating agent for zinc ions in the active site of metalloproteinases, but EDTA can also inhibit other metal ion dependent proteases, such as calcium dependent cysteine proteases.
EDTA may interfere with metal dependent biological processes.

1-2 drops per milliliter of whole blood can be dropped from a 2% EDTA solution as an anticoagulant.
Preparation instructions:
This product slowly dissolves in water at room temperature, up to 0.26M, approximately 96 mg in, with a final volume of 1ml.
The pH value of the solution will be within the range of 4-6.
EDTA salts are more soluble in water as the pH value increases: the more EDTA salts in aqueous solution, the higher the solubility at room temperature.
This can be achieved by gradually adding a concentrated hydroxide sodium solution to the EDTA solution.
Storage/Stability:
A stock solution of 0.5M EDTA in water (pH 8.5) can be stable for several months at 4 ° C. EDTA solution may undergo high-pressure sterilization.
Disodium ethylenediaminetetraacetate explains that chelating agents with divalent cations can inhibit enzyme activity, such as metalloproteinases, because metalloproteinases require the presence of divalent cations to be active.
Preparation of 0.5M EDTA (pH=8.0):
Weigh 186.1g of EDTA-Na2.2H2O186.1g and dissolve it in 800mL of pure water by volume. Add 19g of hydroxide and sodium, then adjust the pH to 8.0 with NaOH and make up to volume. Use after high-pressure sterilization.
Notes:
(1) EDTA-Na2.2H2O is not easily soluble in water and can only be dissolved under alkaline conditions. It can only be completely dissolved when the pH is adjusted to 8.
(2) Pay attention to adjusting the final volume solution pH to 8.0.
