Cell Counting Kit-8 (CCK-8) 利用了Dojindo开发的水溶性四唑盐—WST®-8 (2- (2-甲氧基-4-硝苯基) -3- (4-硝苯 基) -5- (2,4-二磺基苯) -2H-四唑单钠盐),它在电子载体1-Methoxy PMS存在的情况下能够被还原成水溶性的橙黄色甲臜。 CCK-8溶液可以直接加入到细胞样品中,无需配制。CCK-8法是用于测定细胞增殖或毒性实验中活细胞数目的 一种高灵敏度,无放射性的比色检测法。WST®-8被细胞内脱氢酶氧化还原后生成的橙黄色甲臜能够溶解在培养基中 ,生成的甲臜物的数量与活细胞数量成正比。
Cell Counting Kit-8 is a kit used for cell counting in cell proliferation or toxicity experiments such as chemical drugs. The reagent kit uses a highly sensitive new water-soluble tetrazolium salt WST-8 formazane dye as the chromogenic substrate, which significantly improves sensitivity compared to traditional cell counting kits. WST-8 is reduced by intracellular dehydrogenase to produce water-soluble formazane dye. The number of living cells can be measured by directly measuring the absorbance of the methylzan dye (450 nm). The number of cells is directly proportional to the amount of formazine dye. At the same time, the reagent kit is a prepared bottle of solution, which is easy to use.
1) Compared to the [3H] method, no radioactive isotopes are required.
2) The use of water-soluble tetrazolium salts and water-soluble formazane dyes eliminates the need for DMSO liquid exchange procedures. (Required for MTT analysis, etc.)
3) Compared with other water-soluble tetrazolium salt methods (XTT, MTS), it has higher sensitivity and lower toxicity.
4) The reagent kit is packaged in 1 bottle, without the need for advance configuration or preparation of other reagents.
5) The reagent kit has high stability.
6) The reagent can be used simultaneously with phenol red.
