    产品说明：
    活性氧（ROS）是氧正常代谢的天然副产物，在细胞信号传导中起重要作用。然而，在氧化应激相关状态下，ROS水平会急剧增加。ROS的积累导致细胞结构的严重损害。氧化应激在心血管疾病，糖尿病，骨质疏松症，中风，炎症性疾病，许多神经退行性疾病和癌症中的作用已经得到很好的确立。ROS测量将有助于确定氧化应激如何调节不同的细胞内途径。细胞计™荧光 ROS 检测试剂盒使用我们独特的 ROS 传感器来量化活细胞中的 ROS。ROS绿色是细胞渗透性的。当它与ROS反应时，它会产生绿色荧光。该试剂盒是一种优化的“混合和读取”检测形式，与 HTS 液体处理仪器兼容。细胞计™荧光 ROS 检测试剂盒提供灵敏的一步荧光检测，可在孵育一小时后检测活细胞中的细胞内 ROS。该测定可以以方便的 96 孔或 384 孔微量滴定板形式进行，无需分离步骤即可轻松适应自动化。其信号可以使用荧光酶标仪或荧光显微镜轻松读取。

    一目了然
    协议摘要
     
    1. 在生长培养基中制备细胞
    2. 添加 Amplite™ ROS Green 工作溶液（100 孔板为 96 μL/孔，25 孔板为 384 μL/孔）
    3. 将细胞在 37°C 下染色 60 分钟
    4. 用测试化合物处理细胞以诱导ROS
    5. 监测Ex/Em= 490/525 nm（截止= 515 nm）下的荧光增加（底部读数模式）或使用FITC滤光片组的荧光显微镜

    重要事项 在开始实验之前，在室温下解冻所有组分。

    储备液的制备
    除非另有说明，否则所有未使用的储备溶液应分成一次性等分试样，并在制备后储存在-20°C。避免反复冻融循环。
     
    1. Amplite ROS 绿色储备溶液 （500X）：将 40 μL DMSO（组分 C）加入小瓶 Amplite ROS 绿色储备溶液（组分 A）中，充分混合制成 500X Amplite™ ROS 绿色储备溶液。
    ™™避光。注意：20 μL 500X Amplite™ ROS 绿色储备溶液足以用于 1 个板。储存时，请密封管子。

    工作溶液的制备
    将 20 μL 500X Amplite ROS Green 储备溶液加入 10 mL 检测缓冲液（组分 B）中，充分混合制成 Amplite™™ ROS Green 工作溶液。注意：这种Amplite™ ROS绿色工作溶液在室温下至少可稳定保存2小时。

    实验方案样本
    1. 将 100 μL/孔（96 孔板）或 25 μL/孔（384 孔板）的 Amplite™ ROS Green 工作溶液添加到细胞板中。
     
    1. 将细胞在 5% CO 中孵育2，37°C培养箱一小时。
     
    1. 在所需的缓冲液（如PBS或HHBS）中用20 μL 11X测试化合物（96孔板）或10 μL 6X测试化合物（384孔板）处理细胞。对于对照孔（未处理的细胞），加入相应量的复合缓冲液。
     
    1. 为了诱导ROS，在室温或5%CO中孵育细胞板2，37°C培养箱至少15分钟或所需时间（用30mM H处理的Hela细胞为1分钟2O2).
     
    1. 使用荧光酶标仪（底部读取模式）在Ex / Em = 490 / 525 nm（截止= 515 nm）下监测荧光增加，或使用带有FITC滤光片组的荧光显微镜观察细胞。
