    协议摘要
     
    1. 用密度为 5 × 10 的测试化合物制备细胞5至1×106细胞/毫升
    2. 准备并添加ThiolTracTM紫色500细胞工作溶液
    3. 37oC孵化20 到 30 分钟
    4. 读取Ex/Em = 405/525 nm-太平洋橙色滤光片组时的荧光强度

    重要注意事项
    在开始实验之前在室温下解冻。

    储备液的制备
    除非另有说明，否则所有未使用的储备溶液应分成一次性等分试样，并在制备后储存在-20°C。避免反复冻融循环。

    ThiolTracTM紫色 500 储备溶液 （500X）：
    将 200 μL DMSO（组分 C）加入到巯基痕量小瓶中ThiolTracTM紫罗兰色500，搅拌均匀。注意：等分并存储未使用的ThiolTracTM紫罗兰色 500 储备溶液在 -20oC.避免反复冻融循环。

    工作溶液的制备
    ThiolTracTM紫罗兰500工作溶液（1X）：
    加入 2 μL ThiolTracTM紫色 500 储备溶液到 1 mL 检测缓冲液（组分 B）或您选择的缓冲液中，并充分混合。注意： ThiolTracTM紫罗兰500 工作溶液可以在含有血清的细胞培养基中制备。



    实验方案样本
    1. 用测试化合物处理细胞所需的时间。注意：对于贴壁细胞，用0.5mM EDTA轻轻提起细胞以保持细胞完整，并在用ThiolTrace™ Violet工作溶液孵育之前用含血清的培养基洗涤细胞一次。注意：适当的孵育时间取决于所使用的单个细胞类型和细胞浓度。优化每个实验的孵育时间。
     
    1. 以 1000 rpm 离心细胞 4 分钟，然后在您选择的 1 mL 缓冲液中洗涤细胞（可选）。
     
    1. 将细胞重悬于 1 mL ThiolTracTM紫罗兰500工作溶液并在37时孵育oC 培养箱 20 至 30 分钟。
     
    1. 使用太平洋橙色滤光片组（Ex / Em = 405 / 525 nm）的流式细胞仪监测荧光强度。对感兴趣的细胞进行门控，不包括碎片。
