用于RNA提取时DNA的膜上消化以去除DNA污染。
E.Z.N.A. ® RNA抽提试剂盒可以去除大量的基因组DNA污染,对大多数的下游实验,如Northern杂交并不需要特别的处理，但是对于RT-PCR而言，即便是几个分子的核酸片段都可能被检测到。因此RNA中微量的基因组DNA污染对这种灵敏的实验有着非常大的干扰。目前还没有一个抽提试剂盒可彻底去除DNA的污染，除非使用DNase进行处理。
该试剂盒采用无RNase的DNase，专门为膜上DNA酶消化而设计的。在使用E.Z.N.A. ® RNA Kits抽 提RNA时，当RNA结合到硅胶柱的膜.上以后，可以在柱子中加入DNase和特殊的消化缓冲液，孵育酶切DNA，然后再加入洗涤液洗涤去除降解的DNA、DNase和盐，最后洗脱出RNA。使用该方法得到的RNA可确保不存在DNA的污染和DNase的污染。
