Studies in rats reveal that absorption of the drug is much better after intraduodenal administration compared to oral administration. The systemic bioavailability after oral SAMe administration appears to be low. SAMe dose-dependently (12.5, 25, 50, 100 and 200 mg/kg sc.) decreases immobility time in the forced swimming test in mice and rats. Chronic treatment with SAMe is associated with an increase in density of b-receptors, and a decrease in the affinity of a1 adrenoceptors for phenylephrine in rat cerebral membranes.
