谷胱甘肽是一种三肽(g-谷*酰半**酸甘*酸),以还原(GSH)和氧化(GSSG)状态存在。谷胱甘肽过氧化物酶催化GSSG从GSH形成,谷胱甘肽还原酶将GSSG回收为GSH。随着氧化应激水平的增加,细胞内GSSG会积累,GSH / GSSG比率会降低。因此,GSH / GSSG比率的测定是细胞和组织中氧化应激的有用指标。该比色测定基于5',5'-二*代双2-*基*甲酸(DTNB)和GSH之间的反应以形成TNB,其在412nm处表现出最大的吸光度。吸光度的强度与样品中的GSH水平成正比。 Glutathione is a tripeptide (g-glutamylcysteinylglycine) and exists in reduced (GSH) and oxidized (GSSG) states. Glutathione peroxidase catalyzes the formation of GSSG from GSH, and glutathione reductase recycles GSSG to GSH. With increased levels of oxidative stress, intracellular GSSG will accumulate, and the GSH/GSSG ratio will decrease. Therefore, the determination of the GSH/GSSG ratio is a useful indicator of oxidative stress in cells and tissues. This colorimetric assay is based on the reaction between 5’, 5’-Dithiobis 2- nitrobenzoic acid (DTNB) and GSH to form TNB, which exhibits maximum absorbance at 412 nm. The intensity of the absorbance is proportional to the GSH level in the sample.
