MMP-14 (MT1-MMP) 是膜结合的 MMP。当在细胞中表达时，MMP-14 的催化结构域和血红素结合蛋白结构域暴露在细胞表面。这种酶在肿瘤侵袭中起重要作用。520 MMP-14 Assay Kit 使用 5-FAM（荧光团）和 QXL™520（猝灭剂）标记的 FRET 肽底物来连续测量酶活性。在完整的 FRET 肽中，5-FAM 的荧光被 QXL™ 520 淬灭。在 MMP-14 切割 FRET 肽后，5-FAM 的荧光被恢复，并且可以在激发/发射时连续监测 = 490 纳米/520 纳米。具有优越的荧光量子产率和更长的发射波长，检测限：2 ng/ml
MMP-14 diluent: Dilute activated MMP-14 to an appropriate concentration in assay buffer (Component D).
Add test compounds and MMP-14 diluent into a microplate. For one well of a 96-well plate, the suggested volume of enzyme solution is 40uL and 10uL of test compound.
Simultaneously set up the following controls:
Positive control contains MMP-14 diluent without test compound.
Inhibitor control contains MMP-14 diluent and a known MMP-14 inhibitor.
Vehicle control contains MMP-14 diluent and vehicle used in delivering test compound (e.g. DMSO).
Test compound control contains assay buffer and test compound. Some test compounds have strong autofluorescence and may give false results.
Substrate control contains assay buffer.
