Protocol
The cells were fixed with 70% ice-cold methanol overnight at 4℃. Cells stained with Primary Antibody for 30 min at room temperature. The cells were then incubated in 1 X PBS/2%BSA/10% goat serum to block non-specific protein-protein interactions followed by the antibody for 15 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.

有一个简单但很说明问题的实验，可以表明受体交联对于B细胞激活的重要性。分别用抗BCR抗体的Fab片段或F(ab)2片段处理B细胞。Fab因为是单价，不能交联BCR，故不能刺激B细胞活化。F(ab)2可分别与两个BCR分子结合而使之交联，B细胞可有微弱的激活。如果再进一步用抗F(ab)2抗体，通过与两个F(ab)2结合而使更多的BCR交联，则B细胞可获得很强的激活信号。显然，此处实施交联的是抗BCR抗体及抗抗体，而抗原对B细胞激活中发生的是抗原分子或相关复合结构对BCR的交联。 BCR
- 2可使NK细胞增多，诱导K细胞生成，促进活化的B 细胞增殖、分化，提高巨噬细胞的杀伤功能等。因此，IL-2可用于病毒性疾病的治疗，对促使乙肝患者HBsAg，HBeAg转阴等有良好的效果。IL-2 在肿瘤免疫治疗中有重要的地位。研究证明，肿瘤患者体内淋巴细胞产生IL-2的量较正常人明显下降，用高剂量的IL-2治疗肿瘤可获较好疗效，与LAK细胞、TIL细胞联合应用对肿瘤细胞有特异性杀伤活性，而对正常细胞无害。此药国内已进入临床试用阶段，对一些实体瘤，例如肾癌、黑色素瘤和淋巴瘤等有一定疗效。
