Human interleukin family 1, member 7 (hIL1F-7/FIL1- zeta) is a member of the interleukin 1 cytokine family. Five alternatively spliced transcript variants encoding distinct isoforms have been reported with distinct expression profiles. The longest IL1F7 transcript,referred to as IL1F7b or IL1F7 isoform 1, encodes a 218 aa residues proprotein containing a 45 aa propeptide which is cleaved to generate mature protein. IL1F7b binds to IL18 Rα with low affinity but does not exert any IL18 agonistic or antagonistic effects. IL1F7b also binds interleukin 18 binding protein (IL18BP), an inhibitory binding protein of interleukin 18 (IL18), and subsequently forms a complex with IL18 receptor beta subunit,and through which it inhibits the activity of IL18.
·Description Recombinant Human IL1F-7 produced in E. coli (Lys27-Asp192)(accesion# NP_775297), is a single, non-glycosylated, polypeptide chain containing 167 amino acids with an N-terminal Met and having a molecular mass of 18.6kDa.
·Quality Control Biological activity: As measured by its binding ability in a functional ELISA, immobilized rhIL1F7 at 0.1 μg/mL (100 μL/well) can bind rhIL18 R/Fc Chimera with a linear range of 0.015-1μg/mL. Purity: Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Lys-Asn-Leu-Asn. Endotoxin: Less than 0.1ng/μg (IEU/μg) of rHuIL1F-7as determined by LAL test.
·Formulation Sterile and Lyophilized aliquots with a 0.2um filtered protein solution containing 20mM PBS pH7.4.
·Reconstitution We recommend a quick spin followed by reconstitution of rHuIL1F-7 in PBS to a concentration no less than 100ug/ml. This solution can then be further diluted to working solutions.
·Storage Lyophilized rHuIL1F-7 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuIL1F-7 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples with 0.1% HSA or BSA are stable up to 3 months below -18°C for future use.
