Human Chemokine (C-C motif) ligand 8 (CCL8), also known as monocyte chemotactic protein-2 (MCP-2), is produced by human MG63 osteosarcoma cells.CCL8 shares 62% and 58% amino acid identity with MCP-1 and MCP-3 respectively. All three MCP proteins are monocyte chemoattractants. CCL8 is chemotactic for and activates many different immune cells, including mast cells, eosinophils and basophils, which are implicated in allergic response, and monocytes, T cells, and NK cells that are involved in the inflammatory response.CCL8 elicits its effects by binding to several different cell surface receptors including CCR1, CCR2B and CCR5. ·Description Novoprotein recombinant human CCL8/MCP-2 is produced by e.coli transformed with a plasmid contains sequence(Gln24-Pro99) of human CCL8/MCP-2(Uniprot Entry:P80075) fused with a polyhistidine tag at the C-terminus. ·Quality Control Biological activity: The ED50 of human CCL8 as determined by its ability to chemoattract human CCR5 transfected BaF3 mouse proB cells was 0.03-0.1μg/ml. Purity: Greater than 90.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Pro-Asp-Ser-Val.. Endotoxin:Less than 0.1ng/μg (1EU/μg) of CCL8/MCP-2 (Human) ·Formulation The protein was lyophilized after extensive dialysis against PBS, pH 7.4. ·Reconstitution It is recommended to reconstitute the lyophilized rHu-CCL8 in sterile PBS not less than 100 ·Storage Lyophilized rHu-CCL8 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHu-CCL8 should be stored at 4°C between 2-7 days and for future use below -18°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles. |
