Unit Definition One unit of RNase-Free DNase I digests 1 µg of pUC19 DNA to oligodeoxynucleotides in 10 minutes at 37℃. One unit of ScriptGuard RNase Inhibitor results in 50% inhibition of 5 ng of RNase A. Activity is measured by the inhibition of hydrolysis of cyclic 2',3'-CMP by RNase A. Functional Testing The INCOGNITO T7 Ψ-RNA Transcription Kit is functionally tested under standard reaction conditions using the T7 Control Template DNA. The kit must produce at least 90 µg of RNA from 1 µg of the T7 Control Template DNA in 2 hours at 37oC. Contaminating Activity Assays All components of the INCOGNITO T7 Ψ-RNA Transcription Kit are free of detectable RNase and DNase activity, except for the inherent activity of the RNase-Free DNase I component.

CELLSCRIPT™ recently introduced INCOGNITO™ RNA Transcription Kits for in vitrosynthesis of RNA that contains modified nucleosides, such as pseudouridine (Ψ) and/or 5-methylcytidine (m5C) in place of the corresponding U or C canonical nucleosides (Figure 1). These kits are so-named because the capped, polyadenylated and nucleoside-modified RNA products (called "INCOGNITO mRNAs") are disguised so they do not induce innate immune responses to the same extent as the corresponding unmodified mRNAs when transfected into mammalian cells that express a variety of RNA sensors.
