| 描述 | Rotenone is a natural plant-derived insecticide that acts as an inhibitor of mitochondrial electron transport chain complex I. It promotes the generation of mitochondrial reactive oxygen species, induces apoptosis, and is commonly used to establish Parkinson's disease models. |
| 体外活性 | 方法：人白血病细胞 HL-60 用 Rotenone (0-1000 nM) 处理 30 min，使用 Clark oxygen electrode 检测氧气消耗量。 结果：Rotenone 剂量依赖性抑制 HL-60 细胞呼吸。Rotenone 在 500 nM 处抑制细胞呼吸超过 96%。[31] 方法：10th DIV 细胞用 Rotenone (20 nM) 处理 48 h，进行 LDH release assay 检测 LDH 水平。 结果：LDH 的释放可作为一般细胞毒性的指标。Rotenone 诱导 LDH 的释放增加，LDH 活性比对照组增加了 6 倍多。[2] |
