| 描述 | AZD1152 is a pro-drug of Barasertib (AZD1152)-hQPA. Which is a highly selective Aurora B inhibitor with IC50 of 0.37 nM in a cell-free assay. |
| 体外活性 | Barasertib能够抑制诸如HL-60、NB4、MOLM13、PALL-1、PALL-2、MV4-11、EOL-1、THP-1及K562等造血恶性细胞的增殖，其IC50值为3-40 nM，其效力约为另一种Aurora激酶抑制剂ZM334739的100倍左右，ZM334739的IC50为3-30 μM。此外，Barasertib还能以IC50为1 nM及2.8 nM抑制MOLM13和MV4-11细胞的克隆生长，以及1-3 nM的IC50值抑制新鲜分离的伊马替尼耐药白血病细胞，相较于IC50值大于10 nM的骨髓单核细胞，其效果更为显著。Barasertib对Aurora B的选择性>3000倍，对Aurora A的IC50为1.368 μM。Barasertib在KMS12和U266细胞中引起带有4N/8N DNA内容的细胞显著积累，并在KMS18和U266中诱导凋亡。与其他50种丝氨酸-苏氨酸及酪氨酸激酶（包括FLT3、JAK2和Abl）相比，Barasertib的活性更低。Barasertib能够引起带有4N/8N DNA内容的细胞积累，并随着剂量和时间的增加而诱导凋亡。与DEX联合应用时，Barasertib对PMI8226、KMS11和U266细胞生存率的负面影响比单一化合物治疗更为明显。 |
| 体内活性 | 给予Barasertib（25mg/kg）单独使用显著抑制MOLM13异种移植瘤的生长，通过观察到含有吞噬细胞的坏死组织得到确认[1]。Barasertib（25mg/kg/天）治疗使异种移植瘤水平降低，虽然与第一轮治疗后的水平略有下降，但这种下降并不具有统计学意义，表明残余细胞可能对Barasertib的第二轮治疗更具抵抗性[4]。此外，Barasertib（10-150mg/kg/天）以剂量依赖性的方式显著抑制多种人类实体瘤异种移植瘤的生长，包括结肠癌、乳腺癌和肺癌[2]。 |
