| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cattle SPD. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Cattle SPD. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cattle SPD, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Cattle SPD in the samples is then determined by comparing the OD of the samples to the standard curve. |

文献和实验于固相载体的干扰物质。聚苯乙烯等塑料对蛋白质的吸附是普遍性的，而在洗涤时又应把这种非特异性吸附的干扰物质洗涤下来。可以说在ELISA操作中，洗涤是最主要的关键技术，应引起操作者的高度重视，操作者应严格按要求洗涤，不得马虎。 洗涤的方式除某些ELISA仪器配有特殊的自动洗涤仪外，手工操作有浸泡式和流水冲洗式两种，过程如下： （1）浸泡式 a.吸干或甩干孔内反应液；b.用洗涤液过洗一遍（将洗涤液注满板孔后，即甩去）；c.浸泡，即将洗涤液注满板孔，放置1-2分钟，间歇摇动，浸泡时间不可随意缩短；d
