| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human GDF10. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human GDF10. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human GDF10, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human GDF10 in the samples is then determined by comparing the OD of the samples to the standard curve. |

. （六）封闭 封闭(blocking)是继包被之后用高浓度的无关蛋白质溶液再包被的过程。抗原或抗体包被时所用的浓度较低，吸收后固相载体表面尚有未被占据的空隙，封闭就是让大量不相关的蛋白质充填这些空隙，从而排斥在ELISA其后的步骤中干扰物质的再吸附。封闭的手续与包被相类似。最常用的封闭剂是0.05%-0.5%的牛血清白蛋白，也有用10%的小牛血清或1%明胶作为封闭剂的。脱脂奶粉也是一种良好的封闭剂，其最大的特点是价廉，可以高浓度使用（5%）。高质量的速溶食用低脂奶粉即可直接当作封闭剂使用
