检测原理

采用竞争型ELISA：样本中的 PSA 与酶标记的 PSA 竞争结合预包被在微孔板上的抗体。随后加入 HRP‑TMB 底物，酶活性通过比色测定（450 nm）反映样本中 PSA 含量，使用标准曲线进行定量计算。

推荐实验步骤（概览）

1. 稀释样本至适当浓度，加入微孔板并与标准品共孵育（30 min，37 °C）。
2. 洗板 3–5 次（使用 Wash Buffer）。
3. 加入 HRP‑conjugated 抗体，继续孵育 30 min。
4. 再次洗板，加入 TMB 底物显色（约 10–15 min）。
5. 加 Stop Solution 终止反应，立即在酶标仪上读取 450 nm（参考 630 nm）。
6. 使用提供的标准曲线或软件生成定量结果。

Assay Principle

Utilizes a competitive ELISA: PSA in the sample competes with enzyme-labeled PSA for binding to antibodies pre-coated on the microplate. Subsequently, the HRP‑TMB substrate is added, and the enzyme activity, measured colorimetrically at 450 nm, reflects the PSA content in the sample, with quantification calculated using a standard curve.

Recommended Procedure (Overview)

1. Dilute samples to appropriate concentrations, add to the microplate, and incubate together with standards (30 min, 37 °C).
2. Wash the plate 3–5 times (using Wash Buffer).
3. Add HRP‑conjugated antibody and continue incubation for 30 min.
4. Wash the plate again, add TMB substrate for color development (approximately 10–15 min).
5. Add Stop Solution to terminate the reaction, and immediately read the absorbance at 450 nm (reference 630 nm) using a microplate reader.
6. Use the provided standard curve or software to generate quantitative results.

Precautions

• If the Sample Diluent or Assay Buffer shows a slight yellow color or precipitate, it can still be used normally after centrifugation to remove the precipitate.
• Reagents from different lots or different manufacturers must be used separately to avoid result deviations.

• Ensure the plate cover fits well during the incubation stage to prevent sample evaporation.
