检测原理
本试剂盒基于酶偶联反应原理。在反应体系中,3-磷酸甘油酸激酶催化底物生成1,3-二磷酸甘油酸,随后GAPDH逆向催化该产物与NADH反应,生成NAD⁺。通过测定340 nm波长下NADH的吸光度下降速率,即可准确计算出GAPDH的酶活性水平。

Overview
CheKine™ Glyceraldehyde-3-Phosphate Dehydrogenase (GAPDH) Activity Assay Kit (Micro Method) is a highly sensitive colorimetric-based kit designed for the detection of GAPDH enzyme activity in animal and plant tissues, cells, bacteria, culture supernatants, and other liquid samples. As a key enzyme in the glycolytic pathway, the detection of GAPDH activity holds significant research value in metabolism studies, disease mechanism exploration, and drug screening.
Principle
This kit is based on an enzyme-coupled reaction principle. In the reaction system, phosphoglycerate kinase catalyzes the substrate to generate 1,3-bisphosphoglycerate, which is subsequently reversely catalyzed by GAPDH to react with NADH, producing NAD⁺. By measuring the rate of decrease in NADH absorbance at 340 nm, the GAPDH enzyme activity level can be accurately calculated.
