背景介绍(Background):
This protein is a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2014]
R-Spondin 1 (RSPO1) is a secreted signaling molecule that regulates WNT4 expression and is essential for female sex determination. RSPO1 promotes ovarian development by upregulating the canonical WNT/β-catenin signaling pathway to antagonize testis formation. RSPO1 is expressed in the developing ovary, neural tube, kidney, stomach, dermis, mammary glands, limb bud, long bones, craniofacial bones, and vertebrae in the mouse. RSPO1-null mice have decreased fertility and lack duct branching and alveolar formation in the mammary glands, leading to a failure of lactation. Homozygous XX RSPO1 knockout mice show partial sex reversal with masculinized ovaries containing a coelomic blood vessel, fetal oocyte depletion, and ectopic testosterone production. Human mutations in RSPO1 lead to the autosomal recessively inherited disorder palmoplantar hyperkeratosis with squamous cell carcinoma of the skin and 46, XX female-to-male sex reversal. RSPO1 therefore acts as an activator of the canonical signaling pathway in ovarian development.
