描述:甘油是甘油三酯的水解产物。甘油三酯水解生成1分子甘油和3分子游离脂肪酸,称为脂肪分解。脂蛋白酯酶水解血中甘油三酯;胰脂酶水解食物中的甘油三酯;激素敏感脂肪分解酶水解脂肪细胞中的甘油三酯。与游离脂肪酸一样,甘油含量是甘油三酯水解反应的可靠检测指标,但检测更加方便。本试剂盒采用甘油磷酸氧化酶法与经典GPO Trinder酶学反应1,通过比色测定液体样品甘油含量。试剂盒经过优化使得操作简单,最低检测灵敏度10 µmol/L,线性范围10~1200 µmol/L,可靠性和重复性俱佳。适合生物医学、食品实验室检测。
原理:在ATP存在下甘油被甘油激酶磷酸化为3-磷酸甘油,再被甘油磷酸氧化酶氧化产生过氧化氢;在过氧化物酶作用下生色底物,光密度值与甘油浓度成正比。
用途:测定血液、细胞培养基、体液、消化液、各种组织细胞匀浆液、酒类、饮料中的甘油浓度。
